Please note the following clarification to the protocol dated 20 June 2010.
1. A modification to Exclusion criterion 209 was made to align with Exclusion criterion 211. Specifically, per Exclusion criteria 211, the chronic use of low-dose corticosteroid is permitted, however, Exclusion criteria 209 does not specify that the use of low-dose corticosteroids is permitted. As such, Exclusion Criterion 209 was modified to:
"Exclusion Criterion 209: Active autoimmune disease that is not controlled by nonsteroidal anti-inflammatory drugs for active inflammatory disease, including small or large intestine inflammation such as Crohn’s disease or ulcerative colitis, which requires immunosuppressive therapy. Low doses of steroids (equivalent to

Gaucher disease (GD) is characterized by high treatment costs and a significant burden to patients in the form of physical and emotional impairment. Unmet need remains for new high-value treatment options offering security of supply. XXXX is an ERT product for GD which, being derived from carrot cells, is not vulnerable to mammalian viral contamination and provides the reassurance of continuity in supply.
Although GD is rare, the associated burden for patients is significant. Typical manifestations include hepatomegaly, splenomegaly, anemia, and thrombocytopenia, along with skeletal and bone complications. GD weakens the immune systems of sufferers and leads to an increased incidence of peripheral neuropathy, Parkinson’s disease, hematologic malignancies and pancreatic cancer. Physical pain (particularly bone pain) and fatigue, which have a major negative impact on physical well-being, are among the most common symptoms experienced by patients, who also report sadness, anxiety and depression. Crucially, GD patients have a shorter life expectancy than people who do not have the disease. The economic burden of GD is substantial. The annual cost of ERT can be as high as US$600,000 per patient. Besides the burden that it poses to payers, the high cost of treatment is also a burden to patients, a majority of whom report emotional stress relating to the financial aspects of treatment. Due to the treatment cost, the career choices of a significant proportion of GD patients are affected by insurance concerns and coverage.
